We analyzed the transcriptomes of murine B16F10 tongue xenografts from immune-competent C57/B6J mice with or without inhibition of LN-innervating nociceptive nerves using AAV2/retro-Trpv1-hM4D(Gi)-mCherry and Deschloroclozapine. Overall design: The immune-competent C57BL/6J mice underwent retrogradly labeled of LN-innervating nociceptive nerves through injection of eithor AAV2/retro-Trpv1-mCherry or AAV2/retro-Trpv1-hM4D(Gi)-mCherry in cervical LNs. Six weeks later, the B16F10 murine melanoma cells were innoculated into the submucosal region of mouse tongues. Mice were given Deschloroclozapine treatment twice per day for 12 days. The tongue xenografts were harvested and the non-tumoral tissues were removed. The RNA of entire xenografts was extracted and underwent RNA-seq.
