Aiming at the problems that the specific regulatory mechanism of the immune system during the regeneration process is unclear and the correlation between Toll-like receptor(TLR) signaling genes and the regeneration rate is still unclear, this study takes the regeneration model animal, the Mexican salamax, as the research object, aiming to reveal the spatiotemporal expression pattern and function of MyD88-mediated TLR signaling genes during the regeneration process of its tail, to analyze the regulatory effect of this signal pathway on the regeneration process. Using in situ hybridization, real-time quantitative PCR, pharmacological treatments, and cell labeling methods, this study is to reveal the spatiotemporal expression patterns and functions of the MyD88 mediated Toll like receptor(TLR) signaling genes(an important component of the innate immune system) during tail regeneration of axolotl, an important regenerative model animal. The results showed that TLRs and MyD88 were upregulated during the tail regeneration process and were specifically expressed at the regeneration site. After inhibition of the TLR signaling during the regeneration process, it was found that the regeneration speed was faster than that of the normal control group. Furthermore, it was found that inhibiting MyD88 mediated TLR signaling during regeneration promotes cell proliferation, which to some extent explains the accelerated regeneration phenomenon mentioned above. Therefore, MyD88 mediated TLR signaling exhibits a specific dynamic expression pattern, which may play a negative regulatory or balancing role during the regenerative process.
