High detection of Lachnospiraceae members were identified in dysbiotic IL-10 KO mice displaying similar levels of inflammation to control mice. Lachnospiraceae are a highly diverse family of microbes that produce short chain fatty acids, amino acids and antioxidants that may facilitate inflammation relief. Using molecular techniques, five Lachnospiraceae strains were investigated for lowering cellular inflammation and ROS levels. Cell free spent media (CFSM) from Eubacterium rectale resulted in lower ROS, and nitric oxide levels in stressed colon cells. Bioinformatic approaches combined with molecular techniques revealed in E. rectale the potential function of biosynthesis of an antioxidant, glutathione (GSH), and was confirmed using metabolomics on the cell free spent media of E. rectale. To investigate GSH potential for alleviating ROS damage, RNA sequencing of ROS damaged colon cells revealed downregulation of cell stress and immune response genes indicating promotion of recovery and its ability to alleviate ROS stress.
