Perinatal life is a period of extraordinary change in which newborns transition into a new world abundant in environmental stimuli and pathogens. To meet this challenge, the neonatal immune system is equipped with unique functional characteristics and is subject to ongoing development across the early childhood years. To investigate differences in innate immune transcriptional responses, we applied scRNA-Seq to analyze blood collected at birth and 5 years of age following stimulation with two well-characterized model antigens - Lipoloysaccharide (LPS) and Polyinosinic:polycytidylic acid (Poly(I:C)) Matched birth (CBMC) and 5 years (PBMC) blood samples from two donors were treated with LPS and Poly(I:C) treatment, along with matched untreated controls, for 18 hours. Post-culture single cell suspensions were profiled with scRNA-Seq (10X Genomics). ---------------------------- Authors state "The raw sequence data (.fastq) will be made available with controlled access (dbGaP)."
