Heme-TAMs are resistant to tumoricidal transformation by INF?

Inflammatory macrophage activation is a critical component of the antitumor immune response and an emerging therapeutic concept. To evaluate the impact of heme exposure on antitumoral macrophage activity, we grew spheroids of mixed GFP-MC38 and BMDMs that were either untreated or pretreated with heme, INF?, or heme + INF?, and characterized them by scRNA-seq. Overall design: Spheroids of MC38-GFP and MC38-GFP mixed with BMDMs that were stimulated with either IFNy, heme, or heme + IFNy were grown in microwell plates for 9 days. 4500 spheroids per condition were pooled and digested into single cells. Cell Multiplexing Oligos (CMO) labels B301 - B305 (3' CellPlex Kit Set A) were used to label the 5 samples. The 5 samples were pooled and then directly used for GEM generation targeting a cell recovery rate of 10'000 cells.
