T cell receptors of pathogenic CD4 T cells isolated by using distinct phenotypic markers in celiac disease

The main aim of this project is to test in the setting of celiac disease, whether gluten-specific CD4+ T cells may be isolated directly, solely based on this particular phenotype (Tpat) without the use of HLA tetramers (Ttet). We performed highâ€Âthroughput sequencing of rearranged TCRâ€Âα and TCRâ€Âβ genes of Ttet+/Tpat+, Ttet-/Tpat+ and Ttet-/Tpat- CD4+ cells from gut biopsies (exvivo) or that of T cell clones generated from gut biopsies (invitro). The analysis of TCR sequencing data generated from this study will allow us to investigate different features of the TCR repertoire such as clonal expansions, sharing of sequences across donors.EGA study EGAS00001005582
