This PhD research focused on finding new treatment targets for aggressive childhood brain tumours. Using CRISPR gene-editing technology, the study identified genes that are essential for tumour cell survival. A key discovery was the role of cholesterol-producing genes, particularly one called SQLE, which supports tumour growth. Blocking SQLE reduced cancer cell survival, especially under conditions that better mimic real tumours. The study also showed that the environment in which tumour cells are grown can influence research outcomes. These findings highlight the importance of realistic laboratory models and suggest new ways to target childhood brain cancers.
