We developed molecular tool kit to directly measure spatial signal transduction in a model for human neutrophil chemotaxis. Using a light-controllable receptor and a spectrally compatible biosensor for Cdc42 activity, we analyzed the determinants of spatial signal processing. To analyze the role of feedback, we generated a clonal Cdc42 knockout cell line using CRISPR/Cas9. We used amplicon sequencing to analyze the genetic modifications at the Cdc42 locus in this cell line.
