资源简介:

This dataset contains the raw data underlying the figures of the manuscript, as well as the self-developed Matlab scripts used for data analysis in Fig 2B, 3B, 5 and Table 1, and the the project file of the Skyline quantitation used for data analysis in Table S1. Data_Fig1.xlsx: Sheet “citrul” contains the sum of observed cross-links for citrullinated, while sheet “non-citrul” contains data for non-citrullinated samples as presented in Fig. 1 and Table S2. Res A and Res B are the canonical positions of isopeptide bonds in the Protein A and Protein B, respectively. Peptide A and Peptide B columns detail the sequence of the observed peptides. Figure 1 was generated using xiView. Data_FigS1.xlsx: Absorbance values (A340) of forming fibrin clots made of citrullinated and native fibrinogen as measured in time and shown in representative Fig. S1.Data_Fig2A.xlsx: Absorbance values (A340) of 3 or 4 replicates of disassembling fibrin clots made of citrullinated and native fibrinogen +/- FXIIIa (activated factor FXIII) are provided as measured in time and shown in representative figure 2A. Data_Fig2B.zip: Data_Fig2B.xlsx: Absorbance values (A340) of lysing fibrin clots made of citrullinated and native fibrinogen +/- FXIIIa as measured in time (raw data of 3 independent trials on Excel sheets “Raw Data 1-3”). Lysis times (LT10) calculated using the A340-time curves with Matlab script found under LysisCurve_analysing_Fig2.m and evaluated in Fig. 2B and Fig. S2 are available on Excel sheet “LT10”.LysisCurve_analysing_Fig2.m: MATLAB function used for determining lysis time values (LT10) from raw data belonging to Fig. 2B and found on Excel sheet “Raw Data 1-3” of Data_Fig2B.xlsx file.Perform_MW_Fig2.m: MATLAB function used for statistical comparison of datasets shown on Fig. 2B using Mann-Whitney U test. Statistical comparisons are shown on Fig. S2.Data_Fig3B.zip: Perform_MW_Fig3.m: MATLAB function used for statistical comparison of datasets shown on Fig. 3B using Mann-Whitney U test. Statistical comparisons are shown on Fig. S3. Lysis time (LT) values of fibrin clots made of citrullinated and native fibrinogen +/- FXIIIa (raw data as determined by ClotPro device) are also included. Data_Fig4.xlsx: G’ (storage modulus) and G” (loss modulus) values of clotting citrullinated and native fibrinogen +/- FXIIIa as measured in time and shown in Fig. 4A are available on Excel sheet “Kinetic”. Relative deformation (gamma) and dynamic viscosity (eta) values of fibrin clots made from the same fibrinogen samples as a function of stepwise increasing shear stress (Tau) and shown on Fig. 4B and C are available on Excel sheet “FlowCurve”.Data_Fig5.zip: MATLAB function used for statistical comparison of datasets shown on Fig. 5 using Mann-Whitney U test. Maximal clot firmness (MCF) values of fibrin clots made of citrullinated and native fibrinogen +/- FXIIIa (raw data as determined by the ClotPro device) are also included. Statistical comparisons are shown on Fig. S4. Data_Table1.zip: Perform_MW_G1.m: MATLAB script used for the statistical analysis of storage modulus (G′) values of fibrin clots prepared from native and citrullinated fibrinogen, with or without FXIIIa. G′ values as detremined at the end of the clotting phase are included and were compared using the Mann–Whitney U test. The resulting statistical evaluation is presented in Table 1 and Fig. S5.Perform_MW_G2.m: MATLAB script used for the statistical analysis of loss modulus (G”) values of fibrin clots prepared from native and citrullinated fibrinogen, with or without FXIIIa. G’′ values as determined at the end of the clotting phase are included and were compared using the Mann–Whitney U test. The resulting statistical evaluation is presented in Table 1 and Fig. S5.Perform_MW_G2_G1.m: MATLAB script used for the statistical analysis of loss tangent (G’′/G’) values of fibrin clots prepared from native and citrullinated fibrinogen, with or without FXIIIa. G’′/G’ values as determined at the end of the clotting phase are included and were compared using the Mann–Whitney U test. The resulting statistical evaluation is presented in Table 1 and Fig. S7.Perform_MW_gamma.m: MATLAB script used for the statistical analysis of γmax (maximal bearable strain) values of fibrin clots prepared from native and citrullinated fibrinogen, with or without FXIIIa. γmax values as determined right after the clotting phase are included and were compared using the Mann–Whitney U test. The resulting statistical evaluation is presented in Table 1 and Fig. S6.Perform_MW_tau.m: MATLAB script used for the statistical analysis of τ0 (critical shear stress) values of fibrin clots prepared from native and citrullinated fibrinogen, with or without FXIIIa. τ0 values as determined right after the clotting phase are included and were compared using the Mann–Whitney U test. The resulting statistical evaluation is presented in Table 1 and Fig. S6.Data_TableS1.xlsx: Lists the identified citrullinated peptides (sheet “ID”) and the quantity as determined by Skyline (sheet “Quantification”). Area of the peptides is depicted as the sum of all the peptide sequences containing a given citrullination site. Data_TableS1_Skyline.zip: Contains the project file of the Skyline quantitation of data underlying Table S1. Can be opened by Skyline (64-bit) 24.1.0.414.
