We performed single-cell RNA-sequencing analysis on mouse periodontal lesions to analyze the detailed mechanism underlying immune–bone cell interactions in periodontitis. Periodontal tissues were obtained from five control mice and five ligature-induced periodontitis (LIP) mice by stripping the gingival mucosa and scraping the surface of the alveolar bone. After enzymatic digestion, periodontal cells were collected, and single-cell RNA sequencing analysis was performed using the 10x Genomics Chromium system. In this study, data from control and LIP mice were initially combined and analyzed using the integration function, and then separated using the split function to show data from LIP mice.
