Identification of Mettl3-dependent m6A-modified mRNAs upon DNA-damage

The goal of the study was to identify those mRNAs that are m6A-modified in a Mettl3-dependent manner under DNA-damage condition in oncogene-expressing mouse embryonic fibroblasts We performed m6A-eCLIP-seq to identify those Mettl3-dependent m6A RNA modifications occurring after DNA-damage. We utilized oncogene-expressing fibroblasts expressing control shRNAs or shRNAs directed against Mettl3 to acutely knockdown Mettl3, and we treated cells for 6 hours with o,2 ug/ml of doxorubicin before performing m6A-eCLIP-seq.
