Two cell lines derived from 2 Down Syndrome children who developed Acute Lymphoblastic Leukaemia were profiled at the transcriptomic level (bulk RNAseq) and compared against their Patient-Derived Xenograft (PDX) of origin. Overall design: Libraries were prepared from 500 ng of total RNA isoaled from human DS-PER961 (KRAS G12S mutant) and DS-PER-962 (CRLF2 + and JAK2 I682F mutant) cell lines using the SureSelect Stranded PolyA RNASeq kit. Libraries were sequenced at 40 million paired-end reads per sample using a NovaSeq 6000 SP 200 cycles reagent kit (Illumina). Raw data was demultiplexed, underwent quality control, aligned and mapped to the trancriptome (GENECODE v34, GRCh38.p13 assembly).
