DNA methylome analyses and expression analysis for malignant glioma harbouring H3F3A gene mutation

Analysis of methylation status in glioma with H3F3A gene mutation. We performed Whole-genome bisulfite sequencing on a total of 14 clinical samples and a cell line with or without a H3F3A gene mutation, which is a glioma driver gene. A differentially methylated region (DMR) specific for the G34 mutation was identified. ChIP sequencing was performed using anti-H3.3 G34V antibody and anti-H3.3 antibody in a cell line having a G34V mutation, and the relationship between the G34 mutation-specific DMR and the G34V peak was analyzed. We validated G34 mutation-specific methylation pattern using a cell line knocked out the G34V mutation.
