We report that treatment of HeLa cells with 2 hours of 100nM dBET6, which degrades BET bromodomain proteins, leads to global increases of RNA:DNA hybrids (R-loops) and DNA damage (γH2AX). R-loop occupancy was determined using a mutant version of RNase H1 (D210N) which binds to, but cannot resolve, R-loops.
