Transcriptional control of dendritic cell (DC) development has not been fully understood. TRIM33, a transcription co-factor was found to be crucial for transcription regulation during the development of DCs. Genome wide binding site analysis with CUT&Tag revealed co-localization of TRIM33 with CDK9 and Serine 2 phosphorylated RNA polymerase (S2 Pol II) in the common dendritic cell progenitors (CDPs). Overall design: CD11câCD127âCD117intFLT3+ CDPs were sorted by flow cytometry from the bone marrow of wild-type C57BL/6 mice. 1x10^5 cells per sample were subjected to CUT&Tag assay and library construction with the Hyperactive In-Situ ChIP Library Prep Kit purchased from Vazyme under manufactorer's instructions. TRIM33, CDK9, S2 Pol II antibodies , and control IgG were used. Quality-checked libraries were sequenced on an Illumina HiSeq X Ten platform to generate PE150 reads.
