Msh5 is an evolutionarily conserved mismatch repair related protein that promotes meiotic crossovers by stabilizing joint molecule structures such as Holliday junctions. To understand how heterozygosity affects Msh5 binding we used the S. cerevisiae S288c-sp/YJM789 hybrid strain, containing sporulation enhancing QTLs from SK1. Through genome-wide meiotic recombination analysis, we show that the introduction of the QTLs does not impact the meiotic recombination frequency and distribution in S288c-sp/YJM789. To determine Msh5 binding sites we performed ChIP-Seq analysis in the heterozygous yeast model where it is possible to distinguish reads from both the homologous chromosomes using the SNP information. The files submitted contain raw sequence data for the whole genome recombination analysis and the Msh5 ChIP-Seq analysis in the S. cerevisiae S288c-sp/YJM789 hybrid.
