Prostate cancer has the second highest incidence rate in men, yet only an unpredictable 10% of cases progress to metastasis and lethality. Although tumour heterogeneity in primary prostate cancer is a well-established phenomenon, how the subclonal diversity changes, or whether somatic alterations occur during the progression to metastasis is poorly understood. Since mouse models are a very important tool for preclinical modeling, we will address these questions for mouse prostate cancer by sequencing multiple individual metastatic nodules of a highly metastatic mouse model of prostate cancer, driven by loss of Nkx3.1 and Pten, as well as oncogenic mutation of Kras (i.e., Nkx3.1CreERT2/+; Ptenflox/flox;KrasLSL-G12D/+ mice). These studies will complement recent sequencing efforts of human metastatic samples, and help identify the subclones contributing to metastasis as well as the drivers of tumour subclonal expansion.
