<b>T</b><b>etraploid strawberry (</b><b><i>Fragaria moupinensis</i></b><b>)</b>

In order to identify the sex-determining region and gene (s), we sequenced and <i>de novo</i> assembled the female genome of <i>F</i>. <i>moupinensis</i> using 30.80 Gb of PacBio HiFi long reads (34.5X coverage), 151.03 Gb Illumina clean short reads (168.5X coverage), and 43.91 Gb of Hi-C reads (49.1X coverage) . The use of long and accurate HiFi reads coupled with high heterozygosity (5.23%) facilitated the assembly of fully phased haplotypes. This resulted in the generation of four complete haplotypic assemblies named "haplotype A/B/C/D", which were polished for two rounds to produce a tetraploid genome of 896.26 Mb with a contig N50 of 31.68 Mb. Evaluation of k-mers spectra in Smudgeplot revealed a distribution as AAAB (0.58) > AABB (0.13), suggesting the genome of <i>F</i>. <i>moupinensis</i> is autotetraploid . Additionally, the application of Hi-C scaffolding resulted in anchoring 895.13 Mb (99.87%) of the contig sequences to 28 pseudo-chromosomes, with only ten gaps remaining.
