RNA-Seq of mGULL knockdown and hGULL rescue in C57BL6 wild-type mice

For determined h/mGULL have the function similarities, we employed rescue experiments in mice. We knocked down mGULL in mouse liver using shRNAs delivered by adenovirus, and subsequently co-expressed hGULL to assess its capacity to rescue for the loss of function effect of mGULL. After confirmed the mGULL knockdown or hGULL overexpression efficiency, we isolation the total RNA for RNA seq. Seven-weeks old C57BL/6 male were injected via intravitreal injection with adenovirus expressing Control(Lac+pAdv5 virus), shmGULL (shmGULL+pAdv5)or shmGULL+hGULL (for each group, total active viral particles is 5×108) for 7 days before sacrifice.
