It is widely accepted that the expression levels of certain genes undergo significant changes with the cellular growth state. The promoters of these genes may serve as potential candidates for constructing the desired biphasic expression system. Therefore, we performed RNA-Seq analysis of Pichia pastoris during early, mid, and late logarithmic growth phases. Overall design: Logarithmic phase (OD600 value 2-3) of Pichia pastoris GS115 cells (50 µL) were inoculated into 50 mL of YPD medium (1% yeast extract, 2% peptone, and 2% dextrose). The cells were grown under the condition of 30°C with shaking (250-300 rmp). The cells were collected at 24, 36, and 48 hours. Comparative gene expression profiling analysis of RNA-seq data for cells collected at different time points.
