A new hydroxybutanamide derivative from Saussurea involucrata with anti-PD activity on H2O2 insult sH-SY5Y cells

A new hydroxybutanamide derivative, involucramide (1), was obtained from the traditional Chinese medicinal herb Saussurea involucrata. Five other isolates were characterised as betulinic acid (2), costunolide (3), dihydrocostunolide (4), isodihydrocostunolide (5), and dehydrocostus lactone (6). The structure of 1 was established through extensive data analyses encompassing its NMR, IR, UV, and HRESIMS data. All isolates were evaluated for the anti-PD activity on H2O2-insulted SH-SY5Y cells. Amongst them, compound 1 exhibited the potential activity with the EC50 value of 6.84 µM. Furthermore, at the concentration of 20 µM, 1 attenuated the intracellular ROS accumulation, elevated the glutathione (GSH) levels, and reversed the downregulated mRNA expression of HO-1 and NQO1 in H2O2-injured SH-SY5Y cells.
