Regulation of Liver Regeneration by Hepatocyte O-GlcNAcylation in Mice

We studied the role of the post-translational modification called O-GlcNAcylation during liver regeneration. Here we generated O-GlcNAc transferase (OGT-KO) and O-GlcNAcase (OGA-KO) hepatocyte-specific knock-out mice. 70% partial hepatectomy (PHX) was performed to induce liver regeneration. We showed that OGA-KO mice had normal liver regeneration whereas OGT-KO mice had a defect in the termination of liver regeneration. Overall design: We performed RNA-sequencing on 14 and 28 days after PHX on hepatocyte-specific OGT-KO and OGA-KO male, 8 week old mice as well as their respective controls. Equal amounts of RNA was pooled for each sample (n of 3) in each group. Each sample was ran in two different lanes for a targeted 25 million reads.
