Single cell transcriptome sequencing to detect genes in PBMC of patients with systemic lupus erythematosus treated with belimumab. Overall design: This study included three confirmed SLE patients treated with belimumab (SLE_BAFF). According to the 2019 European Union Against Rheumatology (EULAR) and American Society of Rheumatology (ACR) EULAR/ACR criteria, patients diagnosed with systemic lupus erythematosus (SLE) are classified. Patients with immune diseases such as malignant tumors or rheumatoid arthritis were excluded from the study. This study was approved by the Medical Ethics Committee of the First Affiliated Hospital of Shandong Second Medical University. Prior to the test, all participants provided written informed consent and the study complied with the ethical guidelines outlined in the Helsinki Declaration. Collect peripheral blood samples from patients and separate peripheral blood mononuclear cells (PBMCs) using Ficoll Paque gradient centrifugation. Mix three freshly isolated PBMCs and resuspend them in fetal bovine serum with 10% dimethyl sulfoxide. Freeze at -80 ° C for at least 4 hours, store them in liquid nitrogen, and then use them for single-cell transcriptome sequencing (scRNA seq).
