B cell receptor sequences from germinal center (-like) B cells and plasmablasts from inflamed lung and lung-draining lymph node

Germinal center (GC) like B cells can be generated in unstructured non-ectopic T and B cell infiltrates in the in inflamed lung. We used heavy chain B cell receptor (BCR) RNA sequencing to compare antigen-specific B cells from lung to those from the lung-draining lymph node. Antigen-specific B cells from B1-8i and T cells from Smarta mice were adoptively transferred into C57BL/6 recipients, which were repeatedly challenged intranasally with cognate antigen and LPS. On day 18, cells from lung (enzymatically digested) and the tracheal lymph node (Ln) were isolated and further purified for antigen-specific germinal center (like) B cells and plasmablasts.
