Previous data revealed the role of Mfn2 in mediating mitochondrial transfer from a subset of astrocytes that express Dmp1 to cerebrovascular endothelial cells. We used scRNA-seq to investigate the role of Mfn2-mediated mitochondrial transfer in endothelial cell functions. Mfn2 was conditionally depleted in astrocytesDMP1 by establishing Dmp1Cre-Mfn2flox/flox transgenic mice with Dmp1Cre and Mfn2flox/flox mouse lineages. Next, 3 wild type (WT) mouse brains and 3 age-matched Dmp1Cre-Mfn2flox/flox mouse brains were digested into single cells by standard protocols and mixed into one sample, respectively. Mixed single cells from WT mouse brains and Dmp1Cre-Mfn2flox/flox mouse brains were processed for further scRNA sequencing.
