The small intestine is the primary organ for nutrient sensing and absorption, in order to more comprehensively reveal the nutrient sensing and response of different types of intestinal cells, we used single-cell sequencing to reveal the effects of enteral and serosal nutrients on the intestinal tract in a bidirectional nutrient supply mode. To examine the response of different types of cells in the jejunum to the bidirectional nutrition, mice were treated with sham, TPN, or starvation for 24 h, and then 5 cm of jejunal tissue was harvested.To digest the tissues, DNase I (DN25, Sigma-Aldrich) at 1:2000 dilution, Protease Inhibitor Cocktail (HY-K0010, MedChemExpress), Dispase II (40104ES80, Yeasen) and Liberase (5401054001, Roche) at 1:37.5 dilution were added. The tissue was then digested using a gentleMACS Dissociator (Miltenyi Biotec).Library sequencing was performed with the illumina novaseq 6000 pe150.
