Establishment of hematopoietic stem cell compartment after single cell transplantation tracked by FIT approach [scRNA-seq]

Tracing the hematopoietic regeneration of HSC clones by coupling single HSC transplantation and single-cell RNA sequencing (scRNA-seq) of clone-derived cells from 1 to 12 months after transplantation. Overall design: Single HSC from GFP mice accompanied with 5 × 10^5 BM cells (BMCs, competitor cells) from Ly5.2 mice were transplanted into lethally irradiated Ly5.2 mice (4.50 Gy, each of two doses, 3-hour apart) through tail vein injection. Cells were harvested for scRNA-seq from selected repopulated primary recipients after transplanted for 1 month, 4 month and 12 month.
