The goal of this 16S microbiota study was to analyze gut bacterial community structures in nociceptor-ablated mice in different contexts. To ablate nociceptors, three strategies were used. First, Nav1.8-Cre mice were bred with Rosa26-Diptheria toxin (DTA-floxed) mice to generate Nav1.8-Cre/DTA reporter progeny, where Nav1.8-lineage sensory neurons are ablated during development (Cre+ mice) compared with nociceptor-sufficient control littermates (Cre- mice). Second, TRPV1-expressing neurons were chemically ablated by injecting wildtype mice with resiniferatoxin (RTX), a potent TRPV1 ligand that leads to neuronal denervation. Control wildtype mice were injected with vehicle (Veh). Third, TRPV1-Cre mice were bred with Rosa26-Diptheria toxin (DTA-floxed) mice to generate TRPV1-Cre/DTA reporter progeny, where TRPV1-lineage sensory neurons are ablated during development (Cre+ mice) compared with nociceptor-sufficient control littermates (Cre- mice).
