Acute myeloid leukemia remodels the bone marrow non-hematopoietic microenvironment which disrupts niche archiecture and normal hematopoiesis The precise interactions underlying this process are not well understood We used microarrays to detail the global programme of gene expression underlying the interactions between healthy mesenchymal stroma cells and normal hematopoietic progenitors/stem cells (HSPCs) and AML In the first dataset (0611_DoB(5)) MS5 murine mesenchymal stroma cells (MSCs) were cultured either alone or with AML cell lines in myelocult H5100 from STEM CELL TECHNOLOGIES. In the second dataset (0610_DoB(2) and 0910_DoB(2)) MS5 mesenchymal stroma cells were either cultured alone, with human cord blood CD34+ or AML patient samples AML7 and AML8. After 7 days the MSCs were FACS-sorted, followed by RNA extraction and hybridization on Affymetrix microarray.
