On the basis of the clear role of Recombinant human IgG1 in the reduction of liver triglyceride (TG) level elevated by high-fat-diet, the potential effector genes of Recombinant human IgG1 are screened by means of transcriptional techniques. 9 male ApoE-/- mice were randomly divided into 3 groups including control group (N), high-fat-diet group (PBS) and high-fat-diet with Recombinant human IgG1 treatment group (Ab). Briefly, four-weeks-old male ApoE-/- mice were fed a high fat diet( about 4700kcal/kg, 17% of calories from protein, 43% of calories from carbohydrate, 41% of calories from fat ) for 20 weeks and were subsequently transferred to chow food for 1 week, then the mice were treated with intraperitoneal injections of PBS or 3 mg of either Recombinant human IgG1 at age of 25 weeks. After two additional injections at 26 and 27 weeks of age, mice were sacrificed at 29 weeks of age for following analysis. According to the results of liver Oil Red O staining and TG detection, 3 liver samples of each group were detected by trancriptional techniques.
