Puntius denisonii, commonly known as the Denison Barb or Red Line Torpedo Barb, is a freshwater fish species native to the Western Ghats of India. Its unique appearance and vibrant coloration have made it a popular choice in the global aquarium trade. However, over-collection, habitat destruction, and competition from invasive species have led to a significant decline in wild populations, resulting in its classification as Vulnerable in 2009 and Endangered in 2015 by the IUCN Red List. To support conservation efforts, genome sequencing has become essential in understanding the genetic diversity and adaptive traits of P. denisonii. In this study, we present a chromosome-level genome assembly utilizing Illumina short-read, PacBio HiFi, and Hi-C sequencing technologies. The assembled genome spans 736.7 Mb with a contig N50 of 26.80 Mb and a scaffold N50 of 28.66 Mb. Approximately 97.5% of the genome was anchored to 25 chromosomes, including 375.9 Mb of repetitive elements. After masking these elements, we identified 22,569 protein-coding genes. This assembly provides critical insights for phylogenetic studies and conservation strategies, enabling improved management and protection of this endangered species.
