Single-cell gene expression analysis using lung derived cells from SARS-CoV-2-infected mice in aged and adult mice

We conducted single-cell gene expression analysis to elucidate the molecular basis of immune cells in the mouse lung associated with age-dependent severity of COVID-19 infection. In aged infected mice, genes associated with inflammation were more highly expressed in several cell populations compared to adult infected mice. Overall design: We isolated live CD45-positive cells from the lungs of representative mice from aged and adult SARS-CoV-2-infected groups, as well as their uninfected control groups, and subjected them to 10x single-cell gene expression, TCR, and BCR analysis.
