Evaluate heterogeneity of the airway secretory cells, including club secretory, Scgb1a1/Sftpc double positive cells and alveolar secretory cells Overall design: Airways epithelial cells from a double transgenic reporter mouse, Scgb1a1CREERT2/+; Rosa26RAi14/+; Sfptc;GFP/+ were isolated. Labelled cells were isolated using Fluorescence-activated cell sorting (FACS) according to the presence or absence of tdTomato and GFP. We isolated Tomato+ cells (club secretory cells), Tomato/GFP double positive cells (Scgb1a1/Sftpc double positive cells) and GFP positive cells (alveolar secretory cells). Tomato+ cells (club secretory cells) were further separated by FACS, based on podoplanin (Pdpn) expression.
