Re-sequencing targeted regions of chosen human Y chromosomes.. Re-sequencing targeted regions of chosen human Y chromosomes.

We have identified sets of primers that robustly amplify male-specific segments of the human Y chromosome, and can in combination enrich up to several Mb of Y sequence. We and our collaborators are using these to produce pools of amplicons from Y lineages of interest. These pools are indexed and several pools combined for sequencing using Illumina technology. We will discover SNPs and other variants that mark these lineages and use them to refine the human Y phylogeny, and make them available for genotyping in additional samples. Human DNA samples were amplified in Estonia and purified PCR products were sent to WTSI for sequencing.
