Transcription factor induction of vascular blood stem cell niches in vivo [ATAC-Seq 2]

We report accessible chromatin genomic regions using ATAC-sequencing of zebrafish double transgenic for kdrl(flk1):mCherry and sele:GFP from FACS-isolated double positive, single positive, and double negative cells collected from whole embryos at 72 hours post fertilization (hpf). kdrl:mCherry and sele:GFP double positive transgenic embryos were homogenized, filtered, and then up to 50,000 cells from each transgene permutation were FACS sorted into PBS. 12,000-50,000 cells were then lysed and nuclei subjected to Tn5 transposase and libraries were prepared for sequencing.
