To elucidate the sex-specific role of PGC1α in brown adipose tissue (BAT), we performed RNA-seq analysis on BAT from male and female PGC1α knockout mice. Furthermore, to investigate the effects of estrogen on BAT, we conducted RNA-seq analysis on BAT from male and female wild-type mice treated with tamoxifen. experiment 1: PGC-1α knockout experiment Tamoxifen-inducible adipocyte-specific PGC-1α knockout (KO) mice were generated by crossing Ppargc1a flox/flox mice with Adipoq-CreERT2 mice. Eight-week-old male and female KO and Ppargc1a flox/flox control mice were intraperitoneally injected with tamoxifen for 5 consecutive days. BAT samples were collected and analyzed at least 8 weeks after the final injection. The study included four groups based on sex and genotype: Male Control, Male KO (PGC-1α KO), Female Control, Female KO (PGC-1α KO) experiment 2: Tamoxifen treatment experiment In this experiment, eight-week-old male and female wild-type mice were intraperitoneally injected with tamoxifen for 5 consecutive days to evaluate the direct effects of tamoxifen on BAT gene expression. BAT samples were collected and analyzed 2 weeks after the final injection. The study included four groups based on sex and treatment: Male Vehicle (Veh), Male Tamoxifen (TMX), Female Vehicle (Veh), Female Tamoxifen (TMX)
