资源简介:
We previously showed that the RNA of platelet endothelial aggregation receptor-1(PEAR1) is highly expressed at early stages of bovine muscle satellite cells(MDSCs)differentiation. However, its biological function remains unclear. Here, we first utilized western blotting and immunofluorescence techniques to detect the expression of PEAR1 in bovine MDSCs. The expression level of PEAR1 increased between day 0 and day 2 of cell differentiation and decreased from day 3. Moreover, a scratch test showed that the wound healing rate of MDSCs increased after PEAR1 overexpression and decreased upon its suppression. At the same time, we found that, upon PEAR1 induction, both the expression of the focal adhesion-associated and differentiation marker gene, myogenin (MyoG), and the myotube fusion rate increased. However, when the expression of PEAR1 was suppressed, opposite results were obtained. Protein co-immunoprecipitation experiments revealed an association between PEAR1 and integrin beta-1 (ITGB1). Notably, inhibition of focal adhesion kinase (FAK) and ITGB1 repressed bovine MDSC differentiation. In conclusion, our study indicated that PEAR1 is involved in the regulation of bovine MDSC migration and differentiation, and may provide a basis for further exploration of PEAR1 function in bovine muscle development.
