The injury response in the skin is highly complex and involves input from multiple cell types. Single nuclei RNA sequencing was performed to broadly characterize changes in gene expression from all skin cell types early after injury compared to the unwounded state. C57BL/6 mice were used to generate all samples. Unwounded back skin was harvested from two mice for unwounded samples 1 and 2. Full-thickness excisional wounds were made on the back skin of two mice and harvested 1-day post-wound (1dpw) for wounded samples 1 and 2. Nuclei from wound samples and uninjured back skin were isolated with mortar and pestle. Single nuclei library preparations from each sample were generated using 10X Genomics preparation kits.
