Single-cell RNA-sequencing of skin tissues from an acne mouse model topically treated with long-chain saturated fatty acids

Samples were obtained from an acne mouse model in which ethyl palmitate (C16:0), prepared as a 5% (w/w) solution in ethanol, was topically applied to the ears of 7-week-old female C57BL/6J mice. For single-cell RNA-seq analysis, ear tissues were harvested at 0, 6, and 12 hours after application. Single-cell suspensions were generated and subjected to droplet-based single-cell capture, RNA isolation, and library preparation. A total of 9,234, 12,885, and 13,395 cells were analyzed from the 0 h, 6 h, and 12 h samples, respectively. Sequencing was performed using the Illumina NovaSeq 6000 platform.
