TRM cells play an important role in local immune surveillance to prevent pathogen infection and participate in many diseases. But it is not clear whether TRM cells formed in non-lymphoid organs or tissues can mediate the rejection of other allogeneic organ transplants. By transplanting allogeneic skin or heart into SCID mice or Rag2 KO mice, in which induced CD4+ and/or CD8+ TRM cells containing MHC-matched or syngeneic skin was pre-transplanted, we studied With these cells residing in the skin, TRM cells will reject the allograft in the body. RNA-seq analysis and cytology experiments show that during the secondary immune response, the CD4+ TRM cells induced in the skin are conducive to differentiation into Th17-like functional polarization.
