Severe chronic hyperglycaemia (>15mM) causes impaired glycolytic and mitochondrial metabolism in pancreatic ß-cells, leading to dramatically reduced insulin secretion and content. This is associated with dramatic changes in gene expression, in mitochondrial metabolism and in beta-cell function, as described in Haythorne et al (2019) Nature Communications 10, 2474. This study also included bulk RNAseq on islets collected from mice following two weeks of diabetes followed by 2 weeks of euglyceamia (and sequenced at the same time).
