Gain and loss of DNA methylation in cells is a dynamic process that tends to achieve an equilibrium. Many factors are involved in maintaining the balance between DNA methylation and demethylation. The main goal of this work is to discover if Kaiso regulates DNA methylation homeostasis. For this aim, we have used CRISPR/CAS9 genome editing system to introduce a frameshift into Kaiso (ZBTB33) gene in human clear renal carcinoma cells Caki1. Next we performed transcriptome and methylome (WGBS) profilling of Caki1 wildtype and Kaiso-mut cells.
