RNA-seq analysis of RNAs isolated from primary FLT3-ITD+ AML cells in the spleen and bone marrow from xenograft mouse model after 16 hours of treatment with FLT3 inhibitor AC220 (quizartinib) Overall design: FLT3-ITD+ AML cells were engrafted in NRG-S mice. After engraftment (2-5% of leukemic burden in the blood), mice were treated with vehicle or 2.5mg/kg AC220 (quizartinib) for 16 hours, followed by harvest of spleen and bone marrow. Human AML cells were isolated from each compartment. Using RNAs isolated from human AML cells in each compartment, we performed transcriptome analysis.
