Inducing and establishinginduced pluripotent stem cells(iPSCs) can not only explain the mechanism of somatic reprogramming, but also lay a foundation for the establishment of sheep ESCs in vitro.In this study, sheep somatic cells were reprogrammed to sheep iPSCs with stable morphology, pluripotent marker expression and differentiation ability in vitro, delivered by piggyBac transposable with Dox-inducible eight exogenous reprogramming factors. Transcriptome analysis revealed the pluripotent characteristics of somatic reprogramming and insights into sheep iPSCs. The purpose of this study is to establish sheep iPSCs (siPSCs) with more developmental potentialby expressing defined transcription factorsthrough piggyBac+Tet-on transposable system and provided an experimental material for further study on the construction of sheep totipotent embryonic stem cells in the future.
