Triplophysa zhenfengensis belongs to the genus Triplophysa (Teleostei, Nemacheilidae), endemic to Guizhou Province, Southwestern China. The circular mitogenome was 16,564bp in length and consisted of 13 protein-coding genes, two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and one non-coding control region. The overall base composition was 30.61% A, 27.60 % T, 25.46% C, and 16.33 % G with 41.78% GC content. Phylogenetic analysis using mitochondrial genomes of 41 species showed that all Triplophysa species clustered as one monophyletic clade, and T. zhenfengensis was the closest to T. baotianensis. ((T. zhenfengensis+T. baotianensis)+(T. nasobarbatula + (T. xiangxiensis+T. rosa))) make up the first lineage of the genus Triplophysa.
