Bread wheat is an allohexaploid species, and bias in the activities of gene homoeologs is critical for wheat development and trait performance. Asymmetric gene expression of homoeologs was so far investigated in a tissue or organ specific manners, but not at the level of cell types. Here, we performed single nuclei RNA sequencing (snRNA-seq) and single nuclei ATAC sequencing (snATAC-seq) in wheat root to study the asymmetric gene transcription, discover cell differentiation trajectories and cell type specific gene regulatory networks in wheat, for a better understanding of organ development control in this important polyploid crop species.
