Treatment of J-Lat 5a8 cells with KL-2 in combination with the non-canonical NF-kB agonist, AZD5582, induced potent reactivation of HIV-1 transcription. To determine the occupancy of RNA Pol II and CCNT1 at integrated HIV-1 loci following treatment of J-LAT 5a8 cells with KL-2 and AZD5582, RNA Pol II and CCNT1 ChIP-seq was performed. J-Lat 5a8 cells were treated with DMSO, KL-2, AZD5582, and KL-2 + AZD5582 for 48 hours and chromatin was cross-linked with interacting proteins. Proteins of interest (RNA Pol II and CCNT1) were pulled down using specific antibodies and immunoprecipitated DNA was extracted. DNA libraries were prepared and then sequenced.
