This study investigates the role of endothelial ETS1 in regulating cardiomyocyte proliferation and ventricular compact myocardium development. Using ETS1 conditional knockout mice, single-cell RNA sequencing (scRNA-seq) was performed on embryonic day 14.5 (E14.5) ventricles from ETS1 endothelial knockout (eKO) and control mice. Overall design: Single-cell RNA sequencing (scRNA-seq) was performed on embryonic day 14.5 (E14.5) mouse ventricular tissues to investigate the role of endothelial ETS1 in regulating cardiomyocyte proliferation and compact myocardium development. Two ETS1 endothelial knockout samples (Tie2Cre;ETS1^fl/fl) and two control samples (Tie2Cre;ETS1^fl/+) were collected, with each biological replicate consisting of pooled ventricles from three embryos. Single-cell suspensions were prepared by enzymatic dissociation, and libraries were constructed using the Chromium Next GEM Single Cell 3' v3.1 kit (10X Genomics). Libraries were sequenced on the Illumina NextSeq S4 platform.
