We performed V(D)J scRNA sequencing to find transcriptional and B cell receptor repertoire differences in the germinal center of the draining lymph node of mice born to mothers chronically infected with Schistosoma mansoni. Cite-seq was used to help identify antigen specific cells. Overall design: Cells sorted were DAPI-CD19+GL-7+CD95+ from the draining lymph node (popliteal) of mice born to S. mansoni infected and control mothers 14 days after Tetanus/Diphtheria vaccination into the footpad. After sorting, cells were stained with anti-Tetanus toxin-FITC and an anti-FITC oligonucleotide-tagged antibody. Lymph nodes from at least 3 mice were pooled before sorting. Cells were loaded into the 10X Chromium instrument according to the manufacturer's directions. Females from infected mothers were collected and sequenced separately than other samples.
