We report differential gene expression in inflammatory monocyte (IM), tumor-associated macrophage (TAM), and CD8 T cell populations following stereotactic body radiation (SBRT), intratumoral interleukin 12 microsphere (IL-12 MS) delivery, and SBRT/IL-12 MS combination. Immune cell populations were sorted from orthotopic murine pancreatic ductal adenocarcinoma (PDA) tumors prior to RNA sequencing. Overall design: Orthotopic murine PDA tumors (KCKO cell line) were treated with SBRT, IL-12 MS, or SBRT/IL-12 MS combination (duplicate/triplicate); IM, TAM, and CD8 cells were sorted from tumor lysates using a FACSAria II platform; cDNA libraries were constructed using the TruSeq RNA Sample Preparation Kit V2 and sequencing was performed on a HiSeq 2500 platform.
